Comparison of phosphohexose isomerase and lactic dehydrogenase activities in plasma, liver, and tumor tissue of tumor-bearing rats.

چکیده
The mechanisms underlying increases in the activity of various enzymes in the serum or plasma are, in general, incompletely understood. It has been stated that the plasma enzymes are derived from cellular elements either in the tissue or in the blood and that the level of circulating enzymes is therefore an index of the rate of disintegration of such cells (8). On the other hand, in some instances an elevation in activity of a particular plasma or serum enzyme is specifically related to a tissue process involving that enzyme, as, for example, the high serum alkaline phosphatase associated with increased osteoblastic activity in bone or with ob struction of the biliary system. In 1943, Warburg and Christian (19) postulated that the excessive glycolytic activity of tumors might be manifested by increased activity of vari ous glycolytic enzymes in the serum. In pursuance of this concept, several investigations have ex plored the serum levels of aldolase (1, 15-17, 19), phosphohexose isomerase (4—6), and lactic dehydrogenase (2, 10) in animals with transplanted tu mors or in patients with neoplastic disease. Several concepts concerning the factors controlling the level of serum glycolytic enzyme activity have been suggested as a result of those studies (5,6,16) It was believed that further information con cerning these mechanisms might be obtained by comparing the changes occurring in the activities of two glycolytic enzymes in the serum with the changes in the neoplastic tissue, liver, and erythrocytes during the growth of a transplanted

By M. C. BLANCHAER, P. T. GREEN, J. P. MACLEAN AND M. J. HOLLENBERG W IDE ACCEPTANCE of serum glutamic oxaloacetic transaminase estimations1 for the detection of myocardial infarction has stimulated interest in the possible diagnostic uses of other plasma enzymes, including lactic acid dehydrogenase ( LD ) and phosphohexose isomerase ( PHI ) . Both of these enzymes play a role in the intracellu...

Although it has been stated that plasma or serum enzymes are derived from the destruction of cellular elements in the blood or tissues (8), indirect evidence has recently been accumulating that enzymes may pass outward from the cell into the extracellular fluid and thence into the circulation in ways other than those involving necrosis or destruction of the cell (2, 3, 5, 8). It has long been k...
متن کاملPlasma enzymes in tumor-bearing rats.
The plasma enzymes, lactic dehydrogenase, glutamic oxalacetic transaminase, and malic dehydrogenase, increased pro gressively in tumor-bearing rats. The amount of increase varied markedly with the type of tumor and the site of implantation. A lactic dehydrogenase agent could not be demonstrated in any of these animals. Isozyme patterns indicated the tumor was a significant contributor to the in...
متن کاملComparative Biochemistry of Hepatomas. Iii. Carbohydrate Enzymes in Liver Tumors of Different Growth Rates.
The behavior of carbohydrate enzymes was examined in a spectrum of hepatomas of different growth rates, and results were evaluated in the light of data obtained in Hepatoma 5123-D and the Novikoff tumor. The studied biochemical parameters can be classified according to their relation to hepatoma growth rate. There are factors which correlate, others which do not correlate, and, finally, paramet...
متن کاملEnzyme activity, acidic nuclear proteins, and prognosis in human breast cancer.
The activities of the enzymes lactate dehydrogenase, 6-phosphogluconate dehydrogenase, and phosphohexose isomerase in primary human breast cancer biopsies are shown to be related to the time between mastectomy and recurrence of the cancer. These enzymes have higher activity in malignant breast tissues generally than in non-malignant breast tissues. In tumours from patients with long free period...
